PROCEDURE HISTAMINE
1. SAMPLE PREPARATION

How to take a fish sample (clean fish) cut at 3 positions with 1-inch long plate at the back of the pectoral fins, before the pelvic fins and the front of the fish's tail (in the illustration). Then put in the blender to homogenize the sample.

2. EXPLOIT THE SAMPLE EXTRACT AND CHOOSE THE TEST LEVEL

According to research and empirical tests with companies that have used with the Reveal Histamine test we can test at concentrations: 100ppm, 50ppm, 25ppm and 16ppm on the Reveal Histamine test (code: 9501) by changing the procedure. extraction.
- Collect 10 g of homogeneous sample into extraction bottle.
- Add 190ml of distilled water to the extraction sample bottle.
- Cover tightly and shake bottle vigorously for 15 to 20 seconds.
- Wait about 5 minutes, then shake for 15-20 seconds.
- Wait another 5 minutes, and again shake the bottle from 15 to 20 seconds. Allow the tissue to settle to the bottom of the bottle for about 30 seconds. Ready-made samples for diluted extraction (1)

3.EXTRACTION, DILUTE

Because of the sensitivity of the assay, sample extracts must be diluted before performing the test. Using a clean suction tip, add 100 µl of diluted extract (1) to a 7ml (Sample Diluent) vial and close the lid and shake gently. The sample is ready for testing (2). Repeat the operation for other samples with a 7ml (Sample Diluent) vial with each sample. (Attention).

4. PROCESS

- Transfer the sample into the sample cup and place on the stand (or place on a flat position).
- Use a new tip and draw 200ul of the sample above (2) into the cup (3). Repeat for each other test piece with a new tip and a new cup (guaranteed cleaning).
- Place a new histamine test strip with the tip down into the beaker (3).
- Wait for 5 minutes for the sample to react with the test strip (note the time 5-6 minutes).
- Read visual results on a white background or Neogen Reveal AccuScan III Reader.

NOTES:

1 / Samples, when taken, must be stored in plastic or metal tools such as bottles, plastic bags, .. Do not use glassware to hold samples (as recommended by the manufacturer).
